PD-1-IL-2v is a preclinical recombinant fusion protein being developed by PharmaEssentia. It is designed to simultaneously target Programmed Cell Death Protein 1 (PD-1) and the Interleukin-2 (IL-2) receptor. The construct typically utilizes an IL-2 variant (IL-2v) with abolished or reduced binding to the high-affinity IL-2 receptor alpha chain (CD25) to minimize the activation of regulatory T cells (Tregs) and vascular leak syndrome. By tethering the IL-2v to a PD-1-targeting moiety, the drug aims to selectively deliver the cytokine's stimulatory signal to PD-1-expressing, tumor-reactive T cells, thereby enhancing anti-tumor immunity while reducing systemic toxicity. This dual-action approach combines checkpoint inhibition with localized cytokine-mediated immune stimulation.
