Overview

PD-1 inhibitor + sorafenib is a combination therapy regimen being investigated for the treatment of hepatocellular carcinoma (HCC). The regimen consists of a programmed cell death protein 1 (PD-1) inhibitor, which is a monoclonal antibody that blocks the PD-1 checkpoint to restore anti-tumor T-cell activity, and sorafenib, a small-molecule multi-kinase inhibitor. Sorafenib targets several tyrosine kinases, including vascular endothelial growth factor receptors (VEGFR), platelet-derived growth factor receptors (PDGFR), and RAF kinases, thereby inhibiting tumor angiogenesis and cell proliferation. This combination is notably being studied by Guangxi Medical University in Phase 3 clinical trials as an adjuvant therapy to reduce the risk of recurrence in HCC patients following surgical resection, as well as for the treatment of advanced liver cancer. The specific PD-1 inhibitor used in these investigator-initiated trials is often not specified in early trial records, though the class is well-established in oncology.
