Overview

PD-1 inhibitor (Herbert Lyerly) is a preclinical-stage monoclonal antibody being developed by the laboratory of Dr. Herbert Kim Lyerly at Duke University. As a checkpoint inhibitor, it targets the Programmed Cell Death Protein 1 (PD-1) receptor on T cells. The mechanism involves blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), which are often upregulated by tumor cells to evade immune detection. By inhibiting this pathway, the agent releases the inhibitory signaling on T cells, promoting the activation and proliferation of tumor-reactive T cells. This therapeutic strategy is currently being investigated for the treatment of triple-negative breast cancer (TNBC) and metastatic breast cancer.
