Overview

This combination chemo-immunotherapy regimen is being investigated by Sun Yat-sen University Cancer Center for the treatment of relapsed or refractory classical Hodgkin lymphoma (cHL). The regimen integrates a programmed cell death protein 1 (PD-1) inhibitor with the GVD chemotherapy backbone, which consists of gemcitabine, vinorelbine, and liposomal doxorubicin. The PD-1 inhibitor acts as a checkpoint inhibitor to restore T-cell mediated anti-tumor activity by blocking the interaction between PD-1 and its ligands. The chemotherapy components provide complementary cytotoxic effects: gemcitabine inhibits DNA synthesis, vinorelbine disrupts microtubule assembly, and liposomal doxorubicin intercalates into DNA and inhibits topoisomerase II. Clinical research has demonstrated that this combination significantly improves complete remission rates and event-free survival in patients who have failed first-line induction therapy compared to standard GVD chemotherapy alone.
