PD-1 inhibitor + pegaspargase + gemcitabine + oxaliplatin is a combination chemo-immunotherapy regimen being evaluated for the treatment of extranodal natural killer/T-cell lymphoma (ENKTL). The regimen consists of the P-GemOx chemotherapy backbone—comprising pegaspargase (an asparagine-depleting enzyme), gemcitabine (a nucleoside analog), and oxaliplatin (a platinum-based alkylating agent)—combined with a programmed death-1 (PD-1) inhibitor. This therapeutic approach aims to leverage the direct cytotoxic effects of chemotherapy alongside the immune-enhancing properties of checkpoint inhibition to overcome the aggressive nature of ENKTL. It is currently being studied in Phase 2 clinical trials, such as those led by investigator Wei Xu, as a first-line treatment for advanced or non-nasal subtypes of the disease.
