Overview

PD-1 inhibitor + tenofovir + chidamide + lenalidomide is a multi-targeted combination therapeutic regimen being developed by The Affiliated Hospital of Xuzhou Medical University for the treatment of relapsed or refractory Epstein-Barr virus (EBV)-associated lymphoproliferative disorders (LPDs). This synergistic approach integrates four distinct pharmacological strategies: a PD-1 inhibitor to block immune checkpoints and reactivate anti-tumor T-cell responses; tenofovir, an antiviral nucleotide analogue that suppresses EBV replication; chidamide, a selective histone deacetylase (HDAC) inhibitor that epigenetically reactivates latent EBV and enhances tumor cell immunogenicity; and lenalidomide, an immunomodulatory drug (IMiD) that targets the cereblon E3 ubiquitin ligase complex to promote the degradation of specific transcription factors and stimulate immune effector cells. The regimen is currently being evaluated in a Phase 1/2 clinical trial to provide a novel therapeutic option for EBV-driven lymphoid malignancies that are resistant to standard chemotherapy.
