PD-1 knockout antigen-specific TCR-T cells is a gene-edited T-cell receptor-engineered T-cell (TCR-T) therapy being developed by Shenzhen Innovation Immunotechnology for the treatment of melanoma. The therapy involves engineering T cells to express a specific T-cell receptor (TCR) targeting tumor antigens, combined with the CRISPR-mediated knockout of the programmed cell death protein 1 (PD-1) gene. By removing PD-1, the T cells are rendered resistant to PD-L1-mediated immunosuppression within the tumor microenvironment, potentially enhancing their persistence, functional activity, and tumor-killing capacity. The program is currently in the preclinical stage of development.
