Overview

PD-1 knockout engineered T cells are autologous (patient-derived) T lymphocytes that have been genetically modified to lack expression of the programmed cell death protein 1 (PD-1), also known as PDCD-1. This is typically achieved using genome editing technologies such as CRISPR/Cas9. The absence of PD-1 removes a key inhibitory checkpoint on the surface of activated T cells, enhancing their ability to recognize and destroy cancer cells by preventing tumor-induced immune suppression. These modified T cells are expanded ex vivo and reinfused into the patient as a form of adoptive cell therapy. Early clinical studies have shown that this approach is generally well tolerated and may provide disease control in some patients with advanced cancers such as esophageal squamous cell carcinoma[2][3][1].
