PD-1 KO MUC1-targeted CAR-T cells is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Guangzhou Anjie Biomedical Technology Co. Ltd in collaboration with Sun Yat-sen Memorial Hospital. This biologic therapy utilizes patient-derived T cells that are genetically engineered to express a CAR targeting Mucin 1 (MUC1), a tumor-associated antigen overexpressed in various solid tumors. To enhance efficacy and prevent T-cell exhaustion within the immunosuppressive tumor microenvironment, the CRISPR-Cas9 system is employed to knockout the Programmed Cell Death Protein 1 (PD-1) gene. The therapy is currently in Phase I clinical development for advanced solid tumors, including non-small cell lung cancer (NSCLC), esophageal cancer, and breast cancer, where it has demonstrated a favorable safety profile and preliminary signs of clinical activity.
