PD-1 mRNA LNP vaccine is an investigational mRNA-based immunotherapy designed as a checkpoint vaccine. Unlike traditional mRNA vaccines that target tumor-associated antigens, this candidate utilizes lipid nanoparticle (LNP) technology to deliver mRNA encoding the programmed cell death protein 1 (PD-1). By presenting the PD-1 protein as an immunogen, the vaccine aims to stimulate the patient's own immune system to generate high-affinity endogenous antibodies or T-cell responses against the PD-1 receptor. This approach facilitates a form of active checkpoint inhibition, potentially overcoming limitations of exogenous monoclonal antibodies such as the need for frequent dosing and high systemic peaks. It is currently being evaluated for its safety, immunogenicity, and preliminary efficacy in patients with advanced cancers, specifically primary hepatocellular carcinoma (HCC).
