The PD-1 mRNA LNP vaccine is a therapeutic mRNA-based vaccine encapsulated in lipid nanoparticles (LNPs) designed to target the Programmed cell death protein 1 (PD-1) immune checkpoint. Developed by West China Hospital, this vaccine is being investigated for the treatment of advanced primary hepatocellular carcinoma (HCC) in patients who have failed standard-of-care therapies. By delivering mRNA that likely encodes a PD-1 inhibitor or induces an immune response against PD-1-expressing cells, the therapy aims to modulate the PD-1/PD-L1 pathway, thereby restoring or enhancing the anti-tumor activity of T cells within the tumor microenvironment. It is currently being evaluated in a Phase 1/2 clinical trial to assess safety, tolerability, and preliminary anti-tumor efficacy across escalating dose levels.
