Overview

PD-1 non-targeted AttIL-2 is an experimental antibody-cytokine fusion protein designed as a research control for TEV-56278. It consists of an attenuated form of interleukin-2 (AttIL-2) but lacks the PD-1-specific binding domain. In preclinical studies using patient-derived tumor fragments (PDTF) from renal cell carcinoma patients, it is used to differentiate the effects of systemic IL-2 receptor activation from the targeted, localized activation achieved by PD-1-directed delivery. The attenuated IL-2 moiety is engineered to have reduced affinity for IL-2 receptors compared to wild-type IL-2 (aldesleukin), aiming to expand the therapeutic window by reducing systemic toxicity while maintaining anti-tumor activity when properly targeted.
