PD-1 shRNA-modified CD19 CAR-T cells are an investigational autologous cell therapy developed for the treatment of relapsed or refractory B-cell lymphoma. This therapeutic approach involves the genetic modification of a patient's T cells to express a chimeric antigen receptor (CAR) specific for the CD19 antigen, a protein widely expressed on the surface of B-cell malignancies. To enhance the potency and durability of the CAR-T cells, they are further engineered with short hairpin RNA (shRNA) technology designed to knockdown the expression of the programmed cell death protein 1 (PD-1) receptor. By reducing PD-1 levels, these modified CAR-T cells are intended to be less susceptible to exhaustion induced by the PD-1/PD-L1 inhibitory pathway within the tumor microenvironment, potentially leading to improved clinical outcomes compared to conventional CAR-T therapies.
