PD-1 x VEGF x IL-2 trispecific immune modulator is an experimental next-generation immunotherapy developed by WuXi Biologics. This trispecific molecule is designed to overcome the limitations of PD-1 monotherapy by combining three distinct biological mechanisms: PD-1 checkpoint blockade, VEGF-mediated anti-angiogenesis, and targeted IL-2 cytokine signaling. The molecule features a high-affinity anti-PD-1 antibody that directs a proprietary IL-2 mutein specifically to PD-1-expressing, tumor-specific T cells. This IL-2 mutein is engineered with over 1000-fold attenuated binding to IL-2Rβγ and muted CD25 binding to minimize systemic immunotoxicity and the activation of regulatory T cells. Simultaneously, the VEGF-neutralizing component suppresses tumor angiogenesis and modifies the tumor microenvironment to potentiate the efficacy of the PD-1 blockade. Preclinical data indicate that this trispecific approach offers superior safety and efficacy compared to PD-1xVEGF or PD-1xIL-2 bispecific molecules.
