PD-144418 is a **small-molecule, highly selective sigma-1 receptor ligand** developed by Pfizer. In preclinical pharmacology, it showed exceptionally high affinity for the sigma-1 receptor and marked selectivity over sigma-2 and other tested receptors, ion channels, and enzymes. The compound was investigated as a research-stage neuropsychiatric agent with proposed antipsychotic-like properties, including antagonism of mescaline-induced scratching and attenuation of cocaine-induced hyperactivity in mice, but it did not demonstrate anxiolytic or antidepressant activity in the reported models and was not advanced to approved human therapeutic use.
