PD-1Ab21-CAR-T is an engineered chimeric antigen receptor (CAR) T-cell therapy platform designed to secrete a fusion protein, PD-1Ab21, consisting of an anti-PD-1 single-chain antibody fused with interleukin-21 (IL-21). This modification aims to enhance CAR-T cell persistence and anti-tumor efficacy by providing autocrine and paracrine IL-21 signaling, which promotes a memory stem-like phenotype (CCR7+CD62L+CD45RA+TCF-1 high), while simultaneously blocking the PD-1/PD-L1 inhibitory pathway to prevent T-cell exhaustion. Developed by CD(Suzhou)Biopharma and the Chinese Academy of Sciences, this platform has demonstrated superior tumor eradication and cellular expansion in preclinical models of B-cell malignancies (targeting CD19), multiple myeloma (targeting BCMA), and HER2-positive breast cancer compared to conventional CAR-T cells.
