Overview

PD-1Ab21-CD19 CAR-T (also known as BRL-201) is an autologous chimeric antigen receptor (CAR) T-cell therapy engineered to target CD19-positive B-cell malignancies while simultaneously secreting a bifunctional fusion protein (PD-1Ab21) that consists of a PD-1-blocking antibody and Interleukin-21 (IL-21). Developed by BRL Medicine in collaboration with researchers at Soochow University, this therapy is designed to overcome the limitations of conventional CAR-T cells, such as exhaustion and poor persistence, by providing localized checkpoint inhibition and cytokine support within the tumor microenvironment. In Phase 1 clinical trials for relapsed/refractory diffuse large B-cell lymphoma (R/R DLBCL), PD-1Ab21-CD19 CAR-T has demonstrated a manageable safety profile and significant antitumor activity, including high rates of complete metabolic remission and improved durability compared to standard CD19-directed therapies.
