PD-32766 is a novel, highly selective macrocyclic peptide-based radiopharmaceutical that targets carbonic anhydrase IX (CAIX), a cell surface antigen overexpressed in approximately 95% of clear cell renal cell carcinoma (ccRCC) and minimally expressed in normal tissues. Developed by PeptiDream and its subsidiary PDRadiopharma, PD-32766 can be labeled with diagnostic radionuclides such as copper‑64 (^64Cu) for PET imaging or therapeutic radionuclides like actinium‑225 (^225Ac) or lutetium‑177 (^177Lu) for targeted radiotherapy. The agent binds strongly and selectively to CAIX, enabling both tumor-specific imaging and potential therapy (“theranostics”). Early-phase clinical studies have demonstrated high tumor uptake, excellent lesion detection rates on PET/CT, favorable pharmacokinetics with rapid clearance from blood, good tolerability without serious adverse events, and promising anti-tumor effects in preclinical models. The drug is being developed primarily for the diagnosis and treatment of ccRCC but may have broader applications in other CAIX-expressing tumors[1][3][4][5][7][8][9].
