Overview

PD-AFF1, also referred to as AFF1, is an investigational **peptide-based vaccine** designed to target **alpha-synuclein**, a protein implicated in the pathogenesis of **Parkinson's disease (PD)** and other synucleinopathies. The vaccine acts as an **active immunotherapy**, aiming to induce an immune response that facilitates the clearance of pathogenic forms of alpha-synuclein, especially aggregated species, from the brain. Preclinical studies in transgenic mouse models of PD have shown that immunization with PD-AFF1 reduces neurodegeneration, decreases inflammation, and ameliorates both synaptic and memory deficits by decreasing the accumulation of toxic alpha-synuclein aggregates. The therapy also promotes microglial clearance of alpha-synuclein and increases production of anti-inflammatory cytokines, potentially offering disease-modifying benefits in PD and related disorders[1][3][4][7].
