PD-K-OVA is a preclinical polymeric cancer nanovaccine consisting of chicken ovalbumin loaded onto self-assembled nanoparticles of poly(L-phenylalanine)-block-poly(D-lysine). The D-lysine-containing polypeptide nanoparticle functions as a chiral nanoadjuvant that promotes Toll-like receptor 4-associated dendritic-cell maturation, antigen cross-presentation, and OVA-specific adaptive T-cell responses. In mouse B16-OVA melanoma models, PD-K-OVA showed preventive and therapeutic antitumor activity, including a reported 94.9% tumor-inhibition rate that increased to 97.0% when administered with an anti-PD-1 antibody.
