Overview

**PD-L1 CAR T cells** are genetically engineered T lymphocytes designed to express a chimeric antigen receptor (CAR) that specifically recognizes and targets programmed death-ligand 1 (PD-L1) on tumor cells. The CAR is constructed using a single-chain variable fragment (scFv) derived from a PD-L1 monoclonal antibody, fused with co-stimulatory (CD137/CD28) and activation (CD3 zeta) intracellular signaling domains. This design enables the modified T cells to directly identify, bind, and kill PD-L1–expressing tumor cells—most notably in solid tumors such as non-small cell lung cancer. The approach also targets immunosuppressive cells (e.g., myeloid-derived suppressor cells) within the tumor microenvironment which express PD-L1, thereby potentially restoring and enhancing antitumor immune responses. PD-L1 CAR T cells remain investigational, with ongoing phase 1 trials assessing their safety and efficacy in advanced NSCLC and possibly other solid tumors[1].
