Overview

PD-L1-EphA2-CD28 tandem CAR-T cells are a novel dual-targeting chimeric antigen receptor (CAR) T-cell therapy developed by Vaxcell Biotherapeutics for the treatment of solid tumors. The construct utilizes a tandem architecture co-expressing an anti-PD-L1 single-chain variable fragment (scFv) and a monobody-based binder targeting Ephrin type-A receptor 2 (EphA2). By targeting both PD-L1 and EphA2, which are frequently overexpressed in various malignancies, the therapy aims to overcome tumor antigen heterogeneity and the immunosuppressive tumor microenvironment. The CAR construct is delivered via a 3rd generation lentiviral vector and incorporates a CD28 costimulatory domain. Preclinical studies in ovarian, liver, and gastric cancer xenograft models have demonstrated robust anti-tumor activity, superior tumor infiltration, and a favorable safety profile with no significant body weight loss or adverse effects observed at therapeutic doses.
