PD-L1-Fc is a recombinant fusion protein consisting of the extracellular domain of human programmed death-ligand 1 (PD-L1) fused to the Fc region of human immunoglobulin G (IgG). It is designed to mimic the natural ligand for programmed death-1 (PD-1), an inhibitory receptor expressed on T cells. By engaging with PD-1, PD-L1-Fc delivers inhibitory signals that suppress T-cell activation and proliferation. This mechanism has been used in research settings to study immune checkpoint pathways and T cell regulation. Notably, studies have shown that plate-bound or coimmobilized PD-L1-Fc can inhibit T-cell proliferation even independently of direct interaction with CD80 or other binding partners, suggesting a nonspecific mechanism when used in certain assay formats[5]. The primary use of this molecule is as a research tool rather than as a therapeutic agent.
