Overview

PD-L1 inhibitor + apatinib is a combination therapy consisting of an immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1) and the small molecule tyrosine kinase inhibitor apatinib. PD-L1 inhibitors are monoclonal antibodies that block the interaction between PD-1 on T cells and its ligand PD-L1 on tumor cells, thereby enhancing anti-tumor immune responses. Apatinib selectively inhibits vascular endothelial growth factor receptor 2 (VEGFR-2), blocking angiogenesis in tumors. This combination aims to synergistically enhance anti-tumor immunity while inhibiting tumor blood vessel formation. The regimen has been investigated primarily for advanced or metastatic gastric cancer, where it has shown encouraging clinical activity with manageable toxicity profiles in phase II studies.
