**PD-L1-IR700** is an antibody–drug conjugate designed for **near-infrared photoimmunotherapy (NIR-PIT)**. It consists of an anti-PD-L1 monoclonal antibody (such as avelumab or rat IgG2b clone 10F.9G2) conjugated to the photo-absorber dye IRDye700DX (IR700). After systemic administration and accumulation in PD-L1–expressing tumor cells, exposure to near-infrared light activates the IR700 dye, inducing rapid and targeted necrotic cell death of PD-L1–positive cells. Its mechanism allows depletion of both PD-L1–expressing tumor cells and immunosuppressive cells. Preclinical studies in multiple cancer models (lung, ovarian, colorectal) have shown tumor growth suppression and prolonged survival after treatment. Variants of PD-L1-IR700 (such as avelumab-IR700 and RM-0256) are in preclinical or early development by various research groups and companies.
