PD-L1 long1 peptide vaccine is a therapeutic cancer vaccine consisting of a long peptide epitope (approximately 19–23 amino acids) derived from the programmed death ligand 1 (PD-L1) protein. Developed by the Center for Cancer Immune Therapy (CCIT) at Herlev Hospital, Denmark, the vaccine is designed to stimulate the immune system to recognize and eliminate cells expressing PD-L1. Specifically, it induces anti-regulatory T cells (both CD4+ and CD8+) that target PD-L1-expressing tumor cells and immune regulatory cells (such as Tregs and MDSCs) in the tumor microenvironment. By targeting these cells, the vaccine aims to reverse local immunosuppression and enhance the overall anti-tumor immune response. It is typically formulated with the adjuvant Montanide ISA-51 and has been evaluated in clinical trials for metastatic melanoma, basal cell carcinoma, multiple myeloma, and follicular lymphoma, often in combination with other immunotherapies like nivolumab and IDO-derived peptide vaccines.
