PD-L1-silenced dendritic cell vaccine is an autologous cell-based cancer immunotherapy platform. It consists of dendritic cells (DCs) that have been genetically modified (e.g., via siRNA, shRNA, or CRISPR) to silence or knock down the expression of programmed death-ligand 1 (PD-L1), a key immune checkpoint ligand. By silencing PD-L1, these dendritic cells are designed to prevent the inhibition of T-cell activation through the PD-1/PD-L1 pathway during antigen presentation. The vaccine is typically loaded with tumor-associated antigens (such as gastric cancer antigens) to prime and expand tumor-specific cytotoxic T lymphocytes (CTLs), enhancing the anti-tumor immune response. Lanzhou University has investigated this platform in a Phase II clinical trial (ChiCTR1800015690) in combination with cytokine-induced killer (CIK) cell therapy for the postoperative treatment of gastric cancer.
