PD-L1-silenced dendritic cell vaccine + cytokine-induced killer cells is an experimental autologous cellular immunotherapy combination developed by Lanzhou University for the treatment of postoperative gastric cancer. The therapy involves two components: autologous dendritic cells (DCs) in which PD-L1 expression has been silenced (typically via RNA interference) to prevent immune checkpoint-mediated suppression of T-cell activation, and cytokine-induced killer (CIK) cells, which are ex vivo-expanded T lymphocytes (CD3+CD56+) that mediate non-MHC-restricted tumor cell lysis. By reducing PD-L1-mediated inhibition on the antigen-presenting cells, the vaccine aims to more effectively prime and activate tumor-specific T cells, while the CIK cells provide a direct effector population to eliminate residual cancer cells. It is currently being evaluated in a Phase II clinical trial in China.
