This combination immunotherapy regimen is being investigated for the treatment of recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). It consists of three distinct agents: **PD-L1 t-haNK**, an allogeneic natural killer (NK) cell line engineered to express a PD-L1-targeting chimeric antigen receptor (CAR) and a high-affinity CD16 receptor; **N-803 (Anktiva)**, an IL-15 superagonist complex consisting of an IL-15 mutant (IL-15N72D) bound to an IL-15 receptor alpha/IgG1 Fc fusion protein; and **cetuximab (Erbitux)**, a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). The regimen is designed to overcome resistance to prior checkpoint inhibitors by stimulating NK and CD8+ T cell activity via N-803, while PD-L1 t-haNK and cetuximab provide targeted tumor cell killing through CAR-mediated activity and antibody-dependent cellular cytotoxicity (ADCC).
