PD-L1:CD3 T-cell engager is a bispecific therapeutic candidate engineered to target Programmed Death-Ligand 1 (PD-L1) on tumor cells and the CD3 receptor on T-cells. Developed by researchers at the Fred Hutchinson Cancer Center, Seattle Children's Research Institute, and Blaze Bioscience, this T-cell engager (TCE) is specifically designed to address the high unmet need in primary and metastatic central nervous system (CNS) tumors. Preclinical evidence presented at the AACR 2026 Annual Meeting demonstrated that the agent can effectively cross or bypass the blood-brain barrier to eliminate tumors in various intracranial xenograft models, including glioblastoma, diffuse midline glioma, and brain metastases from lung, breast, and colorectal cancers. The drug functions by redirecting T-cell-mediated cytotoxicity to PD-L1-expressing malignant cells. Safety profiles established in non-human primates indicate a manageable toxicity profile at therapeutic dose levels.
