PD-L2 Long201 is a therapeutic cancer vaccine candidate consisting of a 21-amino acid peptide (sequence: SLELQLHQIAALFTVTVPKEL) derived from the programmed death ligand 2 (PD-L2) protein. Developed by the National Center for Cancer Immune Therapy (CCIT-DK) at Herlev Hospital, Denmark, it is designed to induce a T-cell-mediated immune response against cells expressing PD-L2, an immune checkpoint molecule that typically suppresses anti-tumor immunity. In a Phase 1/2 clinical trial (NCT03939234), PD-L2 Long201 was evaluated in combination with a PD-L1-derived peptide and the adjuvant Montanide ISA 51 for the treatment of previously untreated chronic lymphocytic leukemia (CLL) in patients with unmutated IGHV. While the trial demonstrated the vaccine's ability to induce specific immune responses, no formal clinical responses were observed according to iwCLL criteria.
