Overview

PD-SK-375TS is a genetically engineered, tumor cell-specific adapted oncolytic coxsackievirus B3 (CVB3). It was developed through directed viral evolution and genetic modification to enhance its oncolytic activity and safety profile. The virus is designed to specifically target and lyse tumor cells, particularly the colorectal carcinoma cell line Colo320, which is refractory to the parental CVB3 strain, PD-H. The virus has been further modified to include target sites for miR-375 (miR-375TS) in its 3′ UTR, which restricts its replication outside tumors (e.g., in the pancreas and heart), but allows full tumor-specific oncolytic activity. In animal studies, PD-SK-375TS significantly inhibited the growth of Colo320 tumors in xenografted nude mice without causing detectable side effects, unlike the non-adapted parental virus, which was ineffective in this tumor model[1][4].
