Overview

PD1-anti-MUC16 CAR-T cells are a form of chimeric antigen receptor T cell (CAR-T) therapy engineered to target both Programmed cell death protein 1 (PD1) and Mucin 16 (MUC16, also known as CA125). The therapy combines anti-MUC16 single-chain variable fragment (scFv) for tumor recognition with a PD1 component, designed to enhance T cell efficacy in the immunosuppressive tumor microenvironment by engaging both antigens. This dual-target CAR-T cell construct has demonstrated potent cytotoxic activity against MUC16 and/or PDL1-expressing tumor cells (notably in ovarian cancer models), significant cytokine production (e.g., IL-2, IFN-γ, TNF-α), and markedly improved anti-tumor effects in animal models compared with single-target CAR-T cells. Its primary indication is ovarian cancer, with research focused on overcoming resistance in solid tumors[1][2]. This agent represents an advanced cell therapy approach and is an example of next-generation, dual-specificity CAR-T technology.
