Overview

PD1-BCMA-CART is an autologous chimeric antigen receptor T-cell (CAR-T) therapy being developed by Bioray Laboratories for the treatment of relapsed or refractory multiple myeloma. This therapy is distinguished by its use of a non-viral, site-directed integration method (typically utilizing CRISPR/Cas9 or similar gene-editing technology) to insert the CAR construct into a specific genomic locus, specifically the PDCD1 (PD-1) gene. This approach achieves two therapeutic goals simultaneously: the expression of a CAR targeting B-cell maturation antigen (BCMA) on the surface of myeloma cells and the knockout of the PD-1 protein. By eliminating PD-1 expression, the T-cells are designed to be more resistant to exhaustion and immunosuppression within the tumor microenvironment, potentially leading to improved persistence and enhanced anti-tumor efficacy compared to traditional BCMA-targeted CAR-T therapies.
