Overview

PD1 CD19 CAR-T cells refer to a class of next-generation, "armored" chimeric antigen receptor (CAR) T-cell therapies designed to target CD19-expressing B-cell malignancies while simultaneously blocking the PD-1/PD-L1 immunosuppressive pathway. This is typically achieved by genetically engineering the CAR-T cells to co-express a dominant-negative PD-1 (dnPD-1) protein, which acts as a decoy receptor to bind PD-L1/2 without transmitting inhibitory signals, or by using shRNA to downregulate PD-1 expression. A prominent clinical candidate within this platform is ICTCAR014, developed by Innovative Cellular Therapeutics, which has been evaluated in clinical trials (such as ChiCTR1900021295) for the treatment of relapsed or refractory aggressive B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma (DLBCL).
