Overview

PD1 gene knock-down anti-CD19 CAR-T cells is an autologous chimeric antigen receptor (CAR) T-cell therapy engineered to target the CD19 antigen while simultaneously reducing the expression of the Programmed Cell Death Protein 1 (PD-1) receptor through gene knockdown technology. CD19 is a widely expressed surface protein on B-cell lineage cells, making it a primary target for treating B-cell malignancies. The integration of PD-1 knockdown is designed to overcome the inhibitory PD-1/PD-L1 signaling pathway, which often leads to T-cell exhaustion and reduced efficacy in the tumor microenvironment. By silencing PD-1, these CAR-T cells are intended to exhibit enhanced persistence, proliferation, and sustained anti-tumor activity. Developed by The Affiliated Tumor Hospital of Guangxi Medical University, this therapy is being investigated in clinical trials for a broad range of relapsed, refractory, or high-risk B-cell malignancies, including acute lymphoblastic leukemia, various lymphomas, and multiple myeloma.
