Overview

PD1-PSMA-CART is an experimental, autologous CAR-T cell therapy in which patient T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting prostate-specific membrane antigen (PSMA) and are further modified for intrinsic PD-1 blockade. This is achieved through non-viral gene integration, resulting in T cells that both target PSMA-expressing prostate cancer cells and circumvent PD-1-mediated immunosuppression commonly found in the tumor microenvironment. Its mechanism involves direct immunological cytotoxicity to PSMA+ tumor cells and interruption of the PD-1 checkpoint pathway. The primary indication being investigated is refractory castrate-resistant prostate cancer. The therapy is developed by BRL Medicine Inc and clinical evaluation is sponsored by Zhejiang University[4][5].
