PD1-reg-IL2v is a 'switch-on' PD-regulated-IL2v bispecific antibody designed to enhance anti-tumor efficacy while mitigating the systemic toxicity associated with Interleukin 2 (IL-2) therapy. It features a dual-binding antibody arm that can specifically and competitively bind to either PD-1 or an IL-2 variant (IL2v) in a mutually exclusive manner. The IL2v component acts as an IL2Rβγ-biased agonist, preferentially delivering IL-2 signaling through the IL-2Rβγ receptors. The drug's activity is specifically 'switched on' in the presence of PD-1 positive tumor cells, allowing for targeted expansion and differentiation of tumor-antigen-specific T cells into more effective effector cells. This mechanism aims to improve the therapeutic window compared to non-regulated IL2v molecules and standard-of-care anti-PD1 therapy, while also improving pharmacokinetics through peripheral masking.
