Overview

pd1y is a synthetic 20-amino acid peptide derived from the BC loop of the human Programmed Cell Death Protein 1 (PD-1) receptor. It functions as a competitive inhibitor of the PD-1/PD-L1 checkpoint by binding directly to Programmed Death-Ligand 1 (PD-L1), thereby preventing the interaction between PD-1 and PD-L1 that typically suppresses T-cell activation. Primarily utilized as a research reagent and a structural probe, pd1y has demonstrated the ability to restore T-cell proliferation and cytokine production in preclinical models. While it serves as a foundational molecule for the design of more stable peptide mimetics and small-molecule inhibitors in the field of cancer immunotherapy, it has not progressed into formal clinical trials.
