PDE5 siRNA refers to small interfering RNA molecules designed to selectively silence the expression of phosphodiesterase 5 (PDE5). In preclinical research, PDE5 siRNA has been utilized to investigate the role of PDE5 in various pathological conditions, including erectile dysfunction, colorectal cancer, breast cancer, and renal cell carcinoma. By knocking down PDE5 expression, these siRNA molecules prevent the hydrolysis of cyclic guanosine monophosphate (cGMP), leading to cGMP accumulation and subsequent activation of protein kinase G (PKG) signaling. In oncology models, PDE5 knockdown via siRNA has been shown to induce apoptosis, inhibit cell proliferation, and suppress Wnt/beta-catenin-mediated transcription in tumor cells.
