PDK-1 shRNA is a short hairpin RNA (shRNA) designed to silence the expression of pyruvate dehydrogenase kinase 1 (PDK-1). In the context of glioblastoma multiforme (GBM), PDK-1 is often upregulated by hypoxia and contributes to the Warburg effect, where cancer cells prioritize glycolysis over oxidative phosphorylation. By knocking down PDK-1, this therapeutic approach aims to reverse the metabolic phenotype of GBM cells, thereby inhibiting tumor growth, invasion, and the expression of associated oncogenic factors like EGFR and HIF-1α. Research conducted at the University of Illinois College of Medicine at Peoria has demonstrated that PDK-1 knockdown, either alone or in combination with EGFR knockdown or human umbilical cord blood stem cells (hUCBSC), can effectively suppress the malignant progression of GBM.
