PDL1-IFNα is a conditionally active antibody-cytokine fusion protein being developed by Bonum Therapeutics for the treatment of cancer. Utilizing Bonum's proprietary platform, the therapeutic consists of an interferon-alpha (IFNα) effector linked to a monoclonal antibody targeting Programmed cell death 1 ligand 1 (PD-L1). The construct is designed to remain systemically silent in circulation, thereby avoiding the severe toxicities typically associated with recombinant interferon therapy. Upon binding to PD-L1 on the surface of tumor cells, the fusion protein becomes active, concentrating the potent anti-tumor and immunomodulatory effects of IFNα within the tumor microenvironment. This targeted approach aims to stimulate a localized immune response against the tumor while maintaining a favorable safety profile.
