PDL1CAR is an experimental third-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Programmed death-ligand 1 (PD-L1). Developed by researchers at Taipei Medical University and Seattle Children’s Research Institute, the construct utilizes a lentiviral vector to express an anti-PD-L1 single-chain variable fragment (scFv) fused to a composite intracellular signaling domain containing CD28, 4-1BB (CD137), and CD3ζ. By targeting PD-L1, which is often overexpressed on pancreatic ductal adenocarcinoma (PDAC) cells and within the immunosuppressive tumor microenvironment, PDL1CAR T cells aim to overcome immune evasion and T-cell exhaustion. Preclinical data presented at AAI 2020 demonstrated that these engineered T cells could effectively eliminate PD-L1-expressing CFPAC1 cells in vitro and induce significant tumor regression in xenograft mouse models.
