PDM08

PDM08 (also known as PM050489) is a marine-derived small molecule polyketide developed by PharmaMar for the treatment of advanced solid tumors. Originally isolated from the sponge *Lithistida*, PDM08 acts as a potent microtubule inhibitor by binding to tubulin at a site that overlaps with the maytansine-binding site. This binding inhibits microtubule assembly, leading to mitotic arrest in the G2/M phase and subsequent apoptosis of cancer cells. In preclinical studies, PDM08 demonstrated significant antitumoral activity across various murine models, including renal, colon, lung, prostate, and breast cancers. A Phase I clinical trial (NCT01380249) evaluated the safety, pharmacokinetics, and pharmacodynamics of PDM08 administered as an intravenous infusion twice weekly in patients with advanced solid tumors. However, the compound is no longer listed in PharmaMar's active development pipeline, suggesting that its clinical development has been discontinued.
