Overview

pDNA-rIL10 is a **gene therapy modality** utilizing plasmid DNA encoding the recombinant cytokine **interleukin-10 (IL-10)**, an anti-inflammatory protein. It is designed to deliver the IL-10 gene into target tissues via plasmid injection, leading to in situ expression of IL-10 protein for therapeutic benefit. The primary mechanism is local, sustained production of IL-10, which broadly suppresses pro-inflammatory cytokines and modulates immune response, most notably to relieve chronic pain (especially neuropathic pain) and potentially other inflammatory conditions. Delivery methods investigated include intrathecal injection and encapsulation in PLGA (poly(lactic-co-glycolic acid)) microparticles to enhance tissue retention and release. This approach is under preclinical and experimental clinical evaluation for indications like neuropathic pain and osteoarthritis. The plasmid is often delivered without viral vectors, enhancing safety as a non-viral gene therapy.
