PDPHV Plasmid DNA Vaccines is a polyvalent DNA vaccine candidate developed by Worcester HIV Vaccine (University of Massachusetts Medical School) for the prevention of HIV-1 infection. It serves as the priming component of the Polyvalent DNA Prime-Protein Boost HIV Vaccine (PDPHV) strategy. The vaccine consists of five distinct DNA plasmids using the pSW3891 vector: four plasmids encoding codon-optimized gp120 envelope proteins from HIV-1 subtypes A, B, C, and CRF01_AE, and one plasmid encoding the Gag protein from subtype C. Administered via intramuscular injection, the DNA plasmids enter host cells and express these viral antigens, thereby priming the immune system to recognize diverse HIV-1 strains before a subsequent boost with recombinant gp120 proteins. The goal of this regimen is to elicit broad and potent cellular and humoral immune responses against multiple clades of HIV-1.
