PDRIVE-SURVIVIN is a plasmid DNA vector designed for *anticancer therapy*, utilizing a **survivin promoter** to drive expression of an antisense survivin construct[1]. Survivin, a member of the inhibitor-of-apoptosis (IAP) family, is highly expressed in many cancers but absent in most normal tissues. Overexpression of survivin promotes tumor cell survival and induces chemotherapy resistance, making it a promising therapeutic target. PDRIVE-SURVIVIN selectively activates its promoter in survivin-expressing cancer cells, leading to antisense survivin expression, downregulation of survivin and XIAP, induction of autophagy and apoptosis, and inhibition of cancer cell proliferation[1]. Delivery is typically via liposomal formulations for targeted intracellular plasmid delivery. The construct is effective in p53-mutant and wild-type cancer cells, including multidrug-resistant and tamoxifen-resistant lines, with specificity for survivin-expressing cells[1].
