PDV-D is a virus-like protein delivery vehicle platform developed by GenEditBio for the in vivo generation of chimeric antigen receptor (CAR)-T cells. The system encapsulates a complex payload consisting of genome-targeting CRISPR ribonucleoproteins (RNPs) and CAR DNA templates. PDV-D is engineered for T-cell-specific targeting via selective binders and enables site-specific transgene integration downstream of endogenous T-cell-specific promoters, such as the CD3ε locus. This approach aims to provide off-the-shelf, outpatient-applicable CAR-T therapy by bypassing the complexities of ex vivo manufacturing and preconditioning, while reducing the risks of random genomic integration and insertional mutagenesis associated with traditional viral vectors.
