PDX-1 shRNA adenovirus is an adenovirus-mediated gene therapy designed to deliver short hairpin RNA (shRNA) targeting pancreatic and duodenal homeobox 1 (PDX-1). PDX-1 is a homeodomain-containing transcription factor that is normally restricted to pancreatic islets and certain endocrine cells in adults, but is aberrantly overexpressed in various solid tumors, including pancreatic, ovarian, prostate, breast, and liver cancers, where it acts as an oncogene promoting cell proliferation, invasion, and survival. By delivering shRNA via an adenoviral vector, this therapeutic agent silences PDX-1 expression, triggering cancer cell-specific apoptosis and inhibiting tumor growth. It has demonstrated significant anti-tumor efficacy in preclinical in vitro and in vivo models of ovarian, prostate, and pancreatic cancers.
