Overview

PDX-1 siRNA is a small interfering RNA designed to silence the expression of the Pancreatic and duodenal homeobox factor 1 (PDX-1) transcription factor. PDX-1 is a homeodomain-containing protein that is normally restricted to pancreatic islets and certain endocrine cells in adults, but it is frequently overexpressed in various solid human cancers, including pancreatic, breast, prostate, and ovarian malignancies. By utilizing RNA interference (RNAi), PDX-1 siRNA induces apoptosis specifically in PDX-1-positive cancer cells while sparing non-cancerous cells. Preclinical research conducted by institutions such as the UT Health Science Center at San Antonio and CanThera Cancer Therapy Center has demonstrated that silencing PDX-1 with synthetic siRNA or viral-expressed shRNA can significantly inhibit tumor growth in vitro and in vivo xenograft models, suggesting its potential as a universal therapeutic target for PDX-1-expressing cancers.
